Introduction

Burkholderia Glumae is a pathogenic bacterium that is carried by rice seeds, causing panicle Blight (BPB), a disease that can result in significant losses, reaching up to 75% of the total harvest loss. The symptoms of this disease include dark brown to black panicles, standing upright, and empty rice seeds. This study aims to detect the presence of B. Glumae on rice seeds in the Deli Serdang Regency area.

Background

Burkholderia Glumae is a bacterium that is known to cause panicle Blight, a disease that affects rice crops worldwide. The disease is characterized by the formation of dark brown to black panicles, which are often empty and have a characteristic upright appearance. The disease can cause significant losses to rice farmers, with estimates suggesting that up to 75% of the total harvest loss can be attributed to panicle Blight.

Methodology

This study was conducted from March 2022 to December 2022 at the Plant Disease Laboratory, Faculty of Agriculture, University of North Sumatra. The research process began with seed sampling, which was then extracted, isolated, and purified. The sample was then planted on semi-selective media Potato peptone glucose agar (PPGA). The characteristics of bacterial morphology were determined based on the morphology of colonies and bacterial cells, gram staining, pigments, as well as biochemical tests and pathogenicity in onion tubers in vitro.

Results

Based on morphological analysis, biochemistry, and in vitro tests on onion tubers, five isolates were identified as B. Glumae: B.kuis.cih (Var quiz stem. Ciherang), B.kuis.in32 (Var quiz stem. Inpari 32), NR.CIH (Namorambe var. Ciherang), Nr. In32 (Namorambe var. Inpari 32), and Sung.cih (Sunggal var. Ciherang). The isolates were detected from the quiz, namorambe, and sunggal.

Discussion

The discovery of B. Glumae in the Deli Serdang area highlights the importance of strict supervision of the quality of rice seeds used by farmers. With the high level of loss of results that can be caused by this disease, prevention and control steps are needed. Farmers must be aware of the symptoms of this disease and conduct routine tests on the seeds before planting.

In addition, this research becomes an important reference for understanding the spread of pathogenic bacteria in other regions and helps in the development of rice varieties that are resistant to B. Glumae. The existence of innovation in control techniques, such as the use of varieties that are more resistant to disease, as well as the application of good agricultural practices, can help reduce the impact of this disease.

Conclusion

With a focus on early detection and proper management, it is expected to increase rice productivity and the welfare of farmers. Counseling and training to farmers regarding the importance of selection of seeds and disease management must also be increased to minimize losses due to this bacterial disease. The involvement of the government and research institutions in providing technical support and resources is also very crucial in an effort to combat panicle galu disease in rice.
