Introduction

Green beans (Vigna Radiata L.) is one of the most important agricultural commodities in Indonesia, widely cultivated and consumed by the population. The development of new varieties is crucial to meet the increasing demand for this crop. This study was conducted in the pounding of Nibung, Batangkuis, Deli Serdang, North Sumatra, from March to June 2012. The experiment used a random design without factor, with five treatments of colchisin concentration: K0 (0%), K1 (0.04%), K2 (0.08%), K3 (0.12%), and K4 (0.16%). The measurements taken included the percentage of germination, plant height, number of leaves, flowering age, weight of dry pods, number of pods produced, number of seeds in each pod, and weighing one hundred seeds. The data obtained was analyzed using Duncan's Multiple Range Test (DMRT) to test differences between treatments.

Results

The results of this study showed that the application of colchisin has a significant effect on growth, particularly plant height and number of leaves. In addition, there is also a positive impact on production parameters, such as flowering age, weight of dry pods, number of pods produced, and number of seeds in each pod. The results also showed that a higher colchisin concentration (as in K3 and K4) is able to produce better growth, indicating that colchisin can stimulate genetic activity that supports plant growth and development.

Discussion

The use of colchisin as a mutagenic agent in this study shows a great potential in improving the quality of green beans. Increasing the height of the plant and the number of leaves is positively correlated with better photosynthesis capacity, which in turn can increase production results. In addition, production parameters such as the number of pods and seeds also show a good response to the treatment of colchisin. However, it is essential to remember that the use of colchisin must be done carefully, as inappropriate doses can cause negative impacts such as malformations or abnormal growth.

The results of this study provide useful information for farmers and researchers in an effort to increase green bean varieties.
